When you have autoclose set, must the status be set to "Waiting for reply" for it to close? Or can you set it to something like "On Hold", or "In Progress" and have it not autoclose?

The autoclose will only be in effect when the status is "waiting reply (from customer)", any other status will prevent ticket from autoclosing.

All languages, yes, the functionality does not depend on the language.
I did make a mistake in my previous reply though - the status shown in Admin panel must be "Replied" (blue color), meaning staff replied to the ticket but the customer didn't post any new messages.
Try testing it like this:
1. Make sure you have a ticket that has "Status" set to "Replied" - the "Replied" status must show in *admin panel*, not in customer interface!
2. Make sure this ticket was not updated for at least 3 days (last change/updated should be at least 3 days ago)
3. In settings set autoclose tickets to 3 days and save changes
4. Close all browser windows and open HESK admin again - the ticket should be closed.
