Is it possible to automatic assign the ticket to hesk supporter, when he change the ticket status to "In Progress"? - for case when he don't reply, just changing the ticket status. Or he must always manualy assign the ticket to himself by clicking the proper button?

Changing the status and assigning an owner are two separate processes in Hesk, so they need to be done separately.
